利用原位杂交分析几种cyclin和CDK基因在人乳腺癌中的异常表达 被引量:2

摘要 目的探讨细胞周期调控基因cyclin和CDK的异常与乳腺癌发生的相关性。方法采集20例乳腺癌外科手术样品,用原位杂交技术分析了cyclinD1、D3、E、CDK2和CDK4mRNA的表达。结果在大多数乳腺癌样品中,这些基因有一种或几种出现高表达。cyclinD1高表达的样品17例(85%),cyclinD3有12例(60%),cy-clinE有7例(35%),CDK2有6例(30%),CKD4有16例(80%)。cyclinD1和CDK4显色最强,其次是cyclinD3,cy-clinE和CDK2显色较弱,正常乳腺组织为阴性显色。结论推测cyclin和CDK基因,特别是cyclinD1和CDK4的异常表达与乳腺癌的发生有密切关系。 Objective\ Study on the relationship between dysregulated expression of cyclin and CDK genes and tumorigenecity of human breast carcinoma.Methods the expression of cyclin D1,D3,E,CDK2 and CDK4 mRNA of 20 human breast cancer specimens was analyzed.Resuts one or more than one of these genes overexpressed in the majority of the cancer specimens,namely cyclin D1 overexpressed in 17 cancer specimens(85%);cyclin D3 in 12(60%);cyclin E in 7(35%);CDK2 in 6 (30%);CDK4 in 16(80%).The staining intensity of cyclin D1 and CDK4 was strong,cyclin D3 was moderate,cyclin E and CDK2 was weak,the normal breast tissue was negative. Conclusions these data implicate that dysregulated expression of several cyclin and /or CDK genes,particularly cyclin D1 and CDK4,might act as potential factors in the pathogenesis of breast cancer.
