摘要
目的:探讨家系虚寒证与非家系虚寒证在宏观证候及微观基因表达的异同。方法:利用Array Tools和FatiGO等生物信息学软件挖掘基因表达谱数据,获得差异基因、差异功能类及差异分子通路。结果:家系虚寒证和非家系虚寒证在宏观证候和临床表现上没有显著差别,但基因表达水平上显著不同,家系虚寒证和非家系虚寒证差异表达的基因分别有136条和138条,其中只有2条相同,分别为NM003370、NM005252;家系虚寒证组差异功能分布相对集中,主要和能量代谢密切相关,而非家系虚寒证组差异功能分布比较广泛;家系虚寒证的差异通路有3条,而非家系虚寒证组差异通路有8条,二者没有相同的差异通路。结论:家系虚寒证和非家系虚寒证差异表达的基因种类、在染色体上的分布、影响的功能及通路都不相同。对家系虚寒证的基因表达谱分析只能适用于该家系病因分析,对非家系虚寒证的发生机制还需相关的实验研究。但可借鉴研究家系虚寒证基因表达谱的方法。
Objective: To explore the similarities and differences of family and non-family yang-deficiency syndrome in the macro syndrome and micro gene expression.Methods: In order to obtain the different genes,functional classes and molecular pathways,the various data mining methods of bioinformatics in gene expression profile,such as Array Tools and FatiGO were used.Results: The family and non-family yang-deficiency evidence at the macro syndrome and clinical performance was no significant difference,but level of the gene expression was significantly different.Differentially expressed genes of the family and non-family deficiency were respectively 136 and 138,of which only two the same,namely,NM003370,NM005252.Relating to differences function distributed more widely and had eight different channels in non-family yang-deficiency,the function distribution of family yang-deficiency was relative concentration,closely related to energy metabolism,and only had three different channels.Conclusion: The differentially expressed genes between the family and non-family yang-deficiency cards were different effects in the species,the distribution in chromosomes and functions,and played a role through different pathways.The evidence on the family yang-deficiency analysis of gene expression can only be applied to the family cause,the pathogenesis related to yang-deficiency syndrome of non-family need experimental studies.But can learn the method from family yang-deficiency.
