抑癌蛋白维生素D3上调蛋白1在肺癌组织中的表达及意义

Expression and significance of tumor suppressor vitamin D3 up-regulated protein 1 in lung cancer

摘要 目的探讨肿瘤抑制蛋白维生素D3上调蛋白1(vitamin D3 up—regulated protein 1,VDUP1)在肺癌组织中的表达及其与肺癌临床、病理之间的关系和意义。方法应用RTPCR及Western blot分别检测35例肺癌患行的癌组织及癌旁正常组织VDUP1mRNA和蛋白表达水平,进一步按照癌细胞分化程度及肺癌TNM临床分期进行分层比较。结果VDUP1mRNA和蛋白在肺癌组织中表达显著低于癌旁正常组织(P=0.000)。在肺癌原发灶中,VDUP1的表达与TNM分期和细胞分化程度密切相关(P值均〈0.05),但与吸烟、性别、病理类型无关。多重分析结果显示高、中、低分化肺癌中VDUP1的表达量分别为1.17±0.07、0.91±0.26、0.83±0.26,三者间差异具有统计学意义(P=0.023);I、Ⅱ期病例中VDUP1的表达量为1.03±0.20,明显高于Ⅲ、Ⅳ期(0.79±0.24,P=0.004)。结论VDUP1在肺癌中低表达,与肺癌TNM分期及细胞分化有关,可能作为判断肺癌进展和预后的一种有效指标。 Objective To investigate the expression of tumor suppressor vitamin D3 up-regulated protein 1 (VDUP1) in lung cancer, and to reveal its relationship with lung cancer and significance. Methods The expressions of VDUP1 mRNA add protein in lung cancer tissues and adjacent normal lung tissues of 35 patients with lung cancer were detected by real time-polymerase chain reaction and Western blot. The hierarchical comparison was further performed according to cancer cell differentiation and TNM clinical stage of lung cancer. Results The expressions of VDUP1 mRNA and protein in lung cancer tissues were obviously lower than those in adjacent normal lung tissues ( P =0. 000). The expression of VDUP1 protein in lung cancer was related to TNM stage and cell differentiation (all P 〈0.05), but not to smoking, sex, and histological type. Multiple analysis results showed VDUP1 expression was respectively 1. 17±0.07,0.91± 0.26, 0. 83 ± 0.26 in high, middle, and low differentiation lung cancer tissues ( P= 0. 023). VDUP1 expression at stage I -II (1.03±0.20) was significantly higher than that at stage III -IV (0.79±0.24, P =0. 004). Conclusions VDUP1 is lowly expressed in lung cancer, and related with TNM stage of lung cancer and cell differentiation, may be an effective indicator to determine progress and prognosis of lung cancer.
