宫颈癌HeLa细胞抑癌蛋白FBW7调控机制研究

摘要 目的:观察MAPK信号通路阻断剂UO-126对人类宫颈癌细胞株HeLa细胞周期素E(cyclinE)及F-盒和含蛋白7的WD重复结构域FBW7表达的影响。方法:采用RT-PCR、Western blot检测UO-126阻断MAPK信号通路前后cyclin E和FBW7 mRNA及蛋白表达情况。结果:RT-PCR及Western blot结果显示,UO-126作用后HeLa细胞cyclin E mRNA表达水平无明显变化,cyclin E蛋白表达水平明显下降,FBW7 mRNA及蛋白表达水平较未处理HeLa细胞均明显增高。结论:宫颈癌HeLa细胞中FBW7异常表达与MAPK信号通路激活密切相关。UO-126阻断MAPK信号通路可下调cyclin E表达,上调FBW7表达。 Objective:To investigate the effects of UO-126,a MAPK inhibitor,on the expression of cyclin E and F-box and WD repeat domain-containing protein 7(FBW7)of human cervical cancer cell line(HeLa cells).Methods:The mRNA and protein expression of cyclin E and FBW7 were detected by RT-PCR and Western blot before and after MAPK signal was blocked by UO-126,a MAPK inhibitor.Results:RT-PCR and Western blot exhibited that cyclin E mRNA expression had no significant changes and cyclin E protein had significantly decreased after HeLa cells were treated with UO-126,and FBW7 mRNA and protein expression had significantly increased after HeLa cells were treated with UO-126.Conclusion:The abnormal expression of FBW7 is closely related to the activation of MAPK signaling pathway in cervical cancer HeLa cells.Cyclin E is decreased and FBW7 is increased after UO-126 blocks the MAPK signaling pathway.
