摘要
目的研究视黄醇结合蛋白4(RBP4)对胰岛素抵抗大鼠骨骼肌组织磷脂酰肌醇3激酶(PI3K)的影响。方法雄性Wistar大鼠以高脂高糖饲料喂养诱发胰岛素抵抗模型,造模成功后随机分成正常对照(NC)、胰岛素抵抗(IR)和吡格列酮治疗(IR+PIO)三组。喂养8周后处死动物,取血清测血糖(FPG)、胰岛素(FINS)、甘油三酯(TG)、胆固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL),计算胰岛素敏感指数(ISI),ELISA法检测血清RBP4水平,免疫组化方法检测骨骼肌组织中磷脂酰肌醇3激酶(PI3K)表达水平。结果 (1)IR组大鼠RBP4、FINS、LDL、腹部脂肪含量与体重比值高于NC组和IR+PIO组(P<0.01),IR组ISI、HDL低于其他两组(P<0.01),IR+PIO组FBG、TC、TG低于IR组(P<0.01);(2)IR组骨骼肌组织PI3K的表达低于NC组和IR+PIO组(P<0.01);(3)血清RBP4与骨骼肌组织PI3K(r=-0.577,P<0.01)、HDL(r=-0.550,P<0.01)、ISI(r=-0.457,P<0.01)呈负相关,与FINS(r=0.604,P<0.01)、腹部脂肪含量与体重比值(r=0.472,P<0.05)、LDL(r=0.337,P<0.05)呈正相关。结论胰岛素抵抗大鼠血清RBP4升高,吡格列酮可降低其水平。RBP4与大鼠骨骼肌组织PI3K水平负相关,提示其降低胰岛素的敏感性可能与阻碍胰岛素信号转导有关。
Objective To investigate effects of Retinol binding Protein-4(RBP4) on Phosphatidylinositol 3 Kinase(PI3K) in somatic muscle tissue of Insulin resistant rats.Methods We seted up insulin resistant rat model on a high fat diet.Then the rats were divided into normal control group(NC),insulin resistant group(IR) and IR model plus pioglitone therapy group(IR+PIO)at random.After 8 weeks,their blood was extracted for measuring the levels of glucose and serum RBP4,insulin,HDL,LDL,TG,TC.Immunohistochemistry was applied to detect the expression levels of PI3K in Somatic Muscle tissue.Results(1)The RBP4,FINS,LDL,ratio of visceral fat content over body weight,were significantly increased in RI group compared with control or IR+PIO(P0.01);ISI,HDL were significantly lower in IR group compared with control or IR+PIO(P0.01);FBG,TC,TG in IR+PIO group were significantly lower than those in IR group.(2)The expression levels of PI3K of IR group were significantly lower than those of both control group and IR+PIO group in somatic muscle tissue(P0.01);(3)RBP4 was directly coorelated with PI3K in somatic muscle tissue(r=-0.577,P0.01),FINS(r=0.604,P0.01),ratio of visceral fat content over body weight(r=0.472,P0.05),LDL(r=0.337,P0.05),HDL(r=-0.550,P0.01)and ISI(r=-0.457,P0.01).Conclusion The level of RBP4 were increased in IR group and pioglitone could decrease its level.RBP4 was directly coorelated with the levels of PI3K in somatic muscle tissue of IR rats,the mechanisms degrading insulin sensibility might be closely related with the interfere of insulin signal transduction in target tissues.
