摘要
目的:初步探讨点地梅提取物saxifragifolin B体外抗实体瘤活性及其作用机制。方法:Saxifragifo-lin B作用于体外培养的人肝癌细胞BEL-7402、肺癌细胞A549和胃癌细胞SGC7901。光镜下观察肿瘤细胞形态学变化;AO/EB双染法观察细胞死亡;MTT法检测细胞增殖水平,计算IC50;流式细胞术检测细胞凋亡和坏死。结果:Saxifragifolin B诱导3种实体瘤细胞出现凋亡样和坏死样改变。Saxifragifolin B以浓度依赖的方式抑制肿瘤生长,作用24 h IC50分别为13.13μmol/L、9.61μmol/L和11.64μmol/L。Saxifragifolin B可以诱导肿瘤细胞凋亡和坏死。结论:点地梅提取物saxifragifolin B具有抗多种实体瘤的活性,其作用机制可能与诱导细胞凋亡和促进细胞坏死有关。
AIM: To investigate the activity of saxifragifolin B from Androsace umbellata against solid tumors.METHODS: Human hepatoma cell line BEL-7402,human lung adenocarcinoma epithelial cell line A549 and human gastric cancer cell line SGC7901 were treated with saxifragifolin B in vitro.Morphological changes of the tumor cells were observed under microscope.Cell deaths were determined by AO/EB double staining.Cell proliferation was assayed by MTT method.The IC50 values were graphically determined.Apoptosis and necrosis were measured by flow cytometry.RESULTS: Saxifragifolin B induced apoptosis and necrosis in all 3 tested solid tumor cells.Saxifragifolin B inhibited proliferation of tumor cells in a concentration-dependent manner with IC50 values of 13.13 μmol/L,9.61 μmol/L and 11.64 μmol/L at 24 h,respectively.Saxifragifolin B induced both apoptosis and necrosis in the tumor cells.CONCLUSION: Saxifragifolin B from Androsace umbellata has the activity against a variety of solid tumors with the possible mechanisms of inducing both apoptosis and necrosis.
