摘要
目的探讨地塞米松(DEX)对大鼠实验性脑出血后基质金属蛋白酶-9(MMP-9)表达的影响。方法 63只健康雄性Wistar大鼠采用自体血注入法建立右侧基底节区脑出血模型,随机分为A、B、C 3组,A组为DEX干预组;B组为脑出血模型组;C组为假手术对照组,造模成功后每天给予A组DEX(1 ml/kg)腹腔注射,B、C 2组用等量生理盐水替代,分别于造模后6 h、12 h、24 h、3 d、5 d、7 d、14 d 7个时间点取脑,免疫组化检测MMP-9的表达。结果 A组MMP-9表达较B组降低(P<0.05),A、B 2组MMP-9表达较C组升高(P<0.05)。结论 DEX能降低大鼠脑出血后血肿周围脑组织MMP-9的表达,MMP-9表达的变化可能为DEX减轻脑水肿的作用通道之一。
Objective To study the relationship between the expression of matrix metalloproteinase-9(MMP-9) around the hematoma and the application of dexamethasone(DEX) after intra-cerebral hemorrhage(ICH).Methods Sixty-three healthy male Wistar rats were randomly divided into three groups:ICH with DEX treatment group(group A),ICH group(group B) and sham operation group(group C).The model of ICH in right ganglia was established by autologous blood injection.After modeling,the rats of group A were given a daily intra-peritoneal injection of DEX(1ml/kg).Groups of B and C were given equivalent normal saline instead.After intervention,the rats were decapitated respectively,at time points of 6h,12h,24h,3d,7d and 14d.The expression of MMP-9 was measured by immunohistochemistry technique.Results The expression of MMP-9 in group A was lower than that in group B(P0.05).The expression of MMP-9 in groups A and B was higher than that in group C(P0.05).Conclusion The application of DEX can decrease the expression of MMP-9 in the area around hematoma.The change in the expression of MMP-9 may be used as a channel to alleviate cerebral edema.
