解郁舒心片中黄酮类化合物的HPLC和UV法测定 被引量:2

摘要 目的研究解郁舒心片中黄酮类化合物含量的测定方法。方法采用UV法测定总黄酮的含量,HPLC法测定芦丁和金丝桃苷的含量。结果总黄酮在7.5~45.3μg/mL与420nm处的吸收度呈良好的线性关系,平均回收率为100.36%,RSD2.58%。采用shim-packCLC-ODS色谱柱,以水-乙腈-磷酸(825︰175︰1)为流动相,芦丁和金丝桃苷与相邻组分分离良好,分别在0.45~4.50μg,0.06~0.60μg线性关系良好,平均回收率为100.22%,101.22%,RSD为1.74%,1.70%。结论 UV法和HPLC法的结合应用,可对解郁舒心片中黄酮类成分的含量进行有效的控制。 Objective To develop methods for quantitative determination of flavonoids in Jieyushuxin tablets. Methods A UV method was used for determination of total flavonoids and HPLC was used for determination of lutin and hyperoside. Results The linear relationship of the concentrations of total flavonoids and peak areas was good in range of 7.5-5.3 μg/mL with the absorbance at 420nm, the average recovery was 100.36% ( RSD=2.58 ). Lutin and hyperoside were separated well by use of the column shim-pack CLC-ODS and a mixture of water-acetonitrile-phosphate acid ( 825 : 175 : 1 ) as mobile phase. Lutin and hyperoside were in good linearity in range of 0.45-4.50 μ g,0.06-0.60 μ g, the average recoveries were 100.224%, 101.22% (RSD=1.74%, 1.70% ). Conclusion The concentrations of total flavonids, lutin and hyperoside can be in good control with the use of both UV and HPLC.
