摘要
文中对离子交换层析法分离RNA水解液中4种单核苷酸的工艺条件进行了研究。考察了上样pH值、上样量、洗脱剂、洗脱流速等因素对4种核苷酸分离效果的影响。研究确定的最佳分离条件为:核苷酸溶液pH值为2.0,上样量为1.0g/L树脂,洗脱液为蒸馏水,洗脱流速为0.15BV/h。经过2次上柱洗脱可将4种单核苷酸较好分离。
The factors on separation of four kinds of nucleotides from RNA hydrolysates by ion-exchange chromatography, such as sample pH value, sample quantity, eluant, eluting velocity were studied. The optimum technological conditions were determined as follows: sample pH value was 2.0, sample quantity was 1.0g/L ,resin, eluant was distilled water, eluting velocity was 0.15 times as the volume of column per hour. Four kinds of nucleotides were at last separated by two-step column chromatography.
