烟曲霉菌株发酵产壳聚糖酶条件的优化 被引量:2

摘要 通过单因子试验和正交试验对选育的一株产壳聚糖酶菌株UC-233进行发酵条件优化,结果表明其最适氮源是黄豆粕粉,最适碳源是壳聚糖,其最适发酵条件是:黄豆粕粉3.3%,胶体壳聚糖1.5%,培养基初始pH为4.0,250mL三角瓶装液量为50mL,接种量为1mL,发酵温度是30℃,发酵时间为48h,在此条件下菌株发酵液的壳聚糖酶活力达39.5U/mL,是优化前的1.43倍。 Optimization ofUC-233 fermentation conditions was studied via single factor and orthogonal test design. The results showed that: the optimum nitrogen source was soy meal powder, the optimum carbon source was colloid chitosan, the optimized fermentation conditions for chitosanase production were: soy meal powder 3.3%, colloid chitosan 1.5%, initial pH value 4.0,250ml flask containing 50ml medium, inoculation quantity lml, temperatm'e was 35℃, fermentation time was 48h,Under these conditions, the highest chitosanase activity could reach 39.5U/ml, which was 1.43 times compared with that before optimization.
