POK红骨髓性致癌因子和鼠双微基因2蛋白在大鼠肺鳞状细胞癌发生中的表达及意义 被引量:2

摘要 目的探讨POK红骨髓性致癌因子(Pokemon)和鼠双微基因2(MDM 2)蛋白在大鼠肺鳞状细胞癌发生、发展中的关系及意义。方法 90只大鼠分为实验组75只和对照组15只,通过建立大鼠不同阶段肺组织鳞状细胞癌模型,采用免疫组织化学法检测肺鳞状细胞癌、癌前病变及正常肺组织中Pokemon和MDM 2蛋白的表达。结果 Pokemon和MDM 2蛋白从正常组织到鳞状细胞癌呈现逐渐增高的趋势。对照组的Pokemon和MDM 2蛋白表达与增生组比较差别无统计学意义(P>0.05),但低于非典型增生组及鳞状细胞癌组,差别有统计学意义(P<0.05);鳞状细胞癌组的Pokemon和MDM 2蛋白表达明显高于增生组和非典型增生组,差别有统计学意义(P<0.05)。转移癌组的Pokemon和MDM 2蛋白表达高于非转移癌组,差别有统计学意义(P<0.05)。Pokemon与MDM 2蛋白表达呈正相关(r=0.692,χ2=43.080,P<0.05)。结论 Pokemon和MDM 2蛋白在大鼠正常肺组织到肺鳞状细胞癌组织的演变过程中表达逐渐增高,Pokemon和MDM 2蛋白可能共同参与了肺癌的发生、发展和转移过程。 Objective To explore the effect of POK erythroid myeloid ontogenic faceor(Pokemon) and murine double minute 2(MDM 2) protein expression in rat with lung squamous cell carcinoma and its significance.Methods Ninety rats were randomly divided into experiment group(n=75) and control group(n=15).The lung squamous cell carcinoma model of rat at different phases was established,and immunohistochemistry method was taken to detected the Pokemon and MDM 2 protein expression in lung squamous cell carcinoma,precancerous lesions and normal lung tissue respectively.Results Pokemon protein and MDM 2 protein expression showed the trend of gradually increasing form normal tissue to squamous cell carcinoma.There was no statistic significance of Pokemon protein and MDM 2 protein expression between control group and hyperplasia group(P0.05).The Pokemon protein and MDM 2 protein expression in control group were lower than those in hyperplasia group and atypical hyperplasia group(P0.05).The Pokemon protein and MDM 2 protein expression in squamous cell carcinoma group were higher than those in in hyperplasia group and atypical hyperplasia group(P0.05).The Pokemon protein and MDM 2 protein expression in metastatic carcinoma group were higher than those in non-metastatic carcinoma group(P0.05).Pokemon and MDM 2 protein expression was positively correlated(r=0.692,χ2=43.080,P0.05).Conclusion Pokemon protein and MDM 2 protein show gradual rising expressions during the evolution of rat form normal tisssue to lung squamous cell carcinoma.Pokemon and MDM 2 protein might be involved in the development and metastasis of lung squamous cell carcinoma.
