摘要
目的探讨急性髓系白血病(AML)的免疫表型特征与意义。方法采用流式细胞术四色免疫荧光直接标记技术与CD45/SSC设门分析技术,检测105例AML患者,根据抗体积分系统进行分型。结果 105例AML病例中免疫表型分析显示,各种抗原的阳性表达依次为CD13(97.1%)、CD33(90.5%)、MPO(79.0%);干/祖细胞分化抗原表达依次为CD117(77.1%)、HLA-DR(72.4%)、CD34(54.3%)。其中跨系表达:淋系抗原表达阳性的AML(36.4%),主要表达CD19、CD7、CD2、CD10、CD8;跨阶段表达(36.4%):以CD34+CD11b+和CD34+CD11c+共表达2种异常最多见。缺失表达(27.3%):以CD13、CD33及HLA-DR等抗原缺失表达易见。结论流式细胞术四色免疫荧光直接标记法对某些特殊类型的白血病有重要的意义,有助于指导临床诊断、治疗及判断预后。
Objective To study the characteristic of immunophenotype of actue myeloid leukemia(AML) and its significance.Methods Four-color direct immunofluorescence staining methods of flow cytometry and CD45/SSC gating analytical technique were taken for detection of 105 cases of AML,and antibody integration system were used for immunophenotype.Results Immunophenotyping of 105 cases of AML showed that all antigens expressed positive were CD13(97.1%),CD33(90.5%),MPO(79.0%)by turns and stem/progenitor cells differentiation antigen expressed were CD117(77.1%),human leukocyte antigen locus DR(HLA-DR)(72.4%),CD34(54.3%) by turns.Patients with AML accompanied positive lymphocyte antigen expressed CD19,CD7,CD2,CD10,CD8.CD34+CD11b+ and CD34+CD11c+ were the two most common expressed abnormality of bestride the stage.CD13,CD33 and HLA-DR were the most visible in the antigen deletion expressed.Conclusion Four-color direct immunofluorescence staining methods of flow cytometry has important significance for special types of AML,and it can help to guide clinical diagnosis,treatment and prognosis.
