炎症状态下大鼠大网膜Ⅰ型和Ⅱ型乳斑的变化

The changes of omental type Ⅰ and type Ⅱ milky spotsin an inflammatory state.

摘要 目的探讨炎症状态下大鼠大网膜Ⅰ型和Ⅱ型乳斑在数量、面积及巨噬细胞超微结构上的变化。方法实验组SD大鼠腹腔注射沙培林诱发炎症反应,微粒子活性炭和HE染色双重标记大网膜乳斑,Image-ProPlus软件测定乳斑的数量和面积,电镜观察乳斑内巨噬细胞的超微结构。结果与对照组大鼠比较,实验组Ⅰ型乳斑数量显著增多[(11.053±1.543)个/cm2 vs(1.913±0.851)个/cm2,P<0.01],面积显著增大[(0.316±0.066)mm2 vs(0.102±0.041)mm2,P<0.01];Ⅱ型乳斑数量显著增多[(14.147±1.702)个/cm2 vs(9.153±0.818)个/cm2,P<0.01],面积显著增大[(0.486±0.080)mm2 vs(0.421±0.136)mm2,P<0.05];Ⅰ型乳斑数量及面积变化均较Ⅱ型乳斑显著(均P<0.01)。两型乳斑巨噬细胞内的溶酶体丰富,线粒体及粗面内质网肿胀扩张,伪足粗大密集并包裹活性炭颗粒。结论在炎症状态下,两型乳斑数量增多、面积增大,表明两型乳斑在腹腔内均有重要的免疫功能,Ⅰ型乳斑变化更为显著。 Objective To investigate the changes of omental typeⅠ and type Ⅱ milky spots in area, number and uhrastructure of macrophages in inflammatory state of rats. Methods Inflammation was induced by intraperitoneal in- jection of Sapylin in SD rats. Image - Pro Plus software was applied for quantification of activated carbon and HE double - labeled omental milky spots. Uhrastructure of macrophages was observed by electron microscope. Results Significant increase of type type Ⅰ and typeⅡmilky spots in number was observed in inflammatory state [ ( 11. 053 ± 1. 543 ) 个/cm2 vs ( 1. 913 ± 0. 851 ) 个/cm2, P 〈 0. 01 ; ( 14. 147±1. 702) 个/cm2 vs ( 9. 153 ± 0. 818 ) 个/cm2, P 〈 0. 01 ; respectively ) ]. Meanwhile, so was the significant increase in area [ ( 0. 316 ± 0. 066 ) mm2 vs ( 0. 102 ± 0. 041 ) mm2, P 〈 0. 01 ; (0. 486 ± 0. 080) mm2 vs (0. 421 ± 0. 136 ) mm2, P 〈 0. 05 ; respectively ]. Furthermore, significantly more prominent changes in number and area were observed of type I milky spots than those of type Ⅱ milky spots in experimental group ( P 〈 0. 01 ). Rich lysosomes, swelling and expansion of mitochondria and rough endoplasmic reticulum, enlarged pseudopods wrapping carbon particles were observed in macrophages in both milky spots in experimental group. Conclusion Both types of milky spots are increased in number and area under inflammatory status, though more prominent in type I milky spot, suggesting that they play important roles in the peritoneal immunity.
