摘要
目的:通过检测趋化因子CXCL9在结直肠癌中的表达情况,探讨CXCL9与结直肠癌临床病理特征之间的关系。方法:72例结直肠癌患者按Dukes分期分为A、B、C、D 4组,应用Elisa法检测4组及对照组20例志愿者血清CXCL9的含量;应用免疫组织化学方法、Western Blot法检测72例结肠癌组织及癌旁组织中的CXCL9表达,探讨CXCL9与结直肠癌临床病理特征之间的关系。结果:4组结肠癌患者血清CXCL9的平均含量分别为10.21 ng/L1、7.12 ng/L、30.57 ng/L、56.26 ng/L,对照组血清CXCL9平均含量为7.83 ng/L,差异具有统计学意义(P<0.05)。65.3%(47/72)的结肠癌组织CXCL9表达阳性;其中43.1%(31/72)为高表达,56.9%(41/72)为低表达,差异具有统计学意义(P=0.023)。11.1%(8/72)的癌旁组织CXCL9表达阳性,为低表达,与结肠癌比较,差异有统计学意义(P=0.000)。CXCL9的表达与结直肠癌患者的性别、年龄、肿瘤部位、肿瘤大小、组织类型无关(P>0.05);而与肿瘤的分化程度、淋巴转移、Dukes分期有关(P<0.01)。结论:CXCL9在结直肠癌组织中呈较高表达,且表达水平与结直肠癌的浸润、转移等有关。
Objective: To investigate the expression of chemokine CXCL9 in human colorectal cancer and explore its correlation to clinicopathologic features of colorectal cancer.Methods: 72 colorectal cancer patients were divided into four groups(A,B,C,D,respectively).Elisa was performed to detect the concentration of CXCL9 in 72 colorectal cancer patients and 20 volunteers' serum samples;Immunohistochemistry and Western-Blot were also performed to detect the expression of CXCL9 in 72 primary lesions of colorectal cancer and normal colorectal tissues adjacent to cancer lesion,and explore the relationship between the expression of CXCL9 and clinicopathologic features of colorectal cancer.Results: The mean serum concentration of CXCL9 for each group was 10.21ng/L,17.12ng/L,30.57ng/L,56.26ng/L and 7.83ng/L respectively(P0.05).Analysis of CXCL9 expression in the 72 colorectal cancer tissues revealed that 65.3%(47/72)of specimens showed positive staining for CXCL9,with 43.1%(31/72)demonstrating strong(2+ and 3+)intensity and 56.9%(41/72)low(-and +)intensity(P=0.023);However,11.1%(8/72) normal colorectal tissues showed low positive staining for CXCL9(P=0.000).CXCL9 expression was closely related to histological differentiation,lymph node metastasis,Dukes stage(P0.01),but no significant differences were observed regarding gender,age,tumor location,size,histological type(P0.05).Conclusion: The expression of CXCL9 in human colorectal cancer is up-regulated,and the level of CXCL9 is correlated to progression and metastasis of colorectal cancer.
