摘要
目的研究机械应力刺激对成牙骨质细胞矿化相关基因骨涎蛋白(bone sialoprotein,BSP)mRNA表达的影响。方法利用四点弯曲细胞力学加载装置,以体外培养的成牙骨质样细胞OCCM-30为研究对象,运用实时荧光定量技术,比较2 000、4 000μstrain张应力和压应力加载组和不加力组OCCM-30在3 h、6 h、12 h、24 hBSP mRNA表达差异。结果和对照组相比,2 000、4 000μstrain的张应力和压应力在4个时间点均抑制OCCM-30 BSP mRNA的表达,且差异有统计学意义(F=5.614,P<0.05)。结论成牙骨质细胞是力学敏感性细胞,机械应力刺激通过影响其BSP mRNA的表达,进而可能影响成牙骨质细胞在牙根吸收和修复中的作用。
Objective To evaluate the effects of the mechanical stress on the express of the mineral associated gene BSP in cementoblast in vitro.Methods Cementoblasts OCCM30 were subjected to mechanical stress by four-points bending system with tension and compression stress at 2 000 μand 4 000 μ for3 h,6 h,12 h,24 h.The expression of BSP mRNA were analysed with real time quantitive RT-PCR after mechanical loading.Results BSP mRNA was suppressed by the stresses.There is a significant difference between subjects and controls(F=5.614,P0.05).Conclusion Cementoblasts are sensitive to mechanical stress,and may play a role in regulating orthodontic-related root resorption or repair.
