摘要
目的:观察银杏叶提取物对糖尿病大鼠肾组织中基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)表达的影响。方法:取成年SD级大鼠50只,随机分成3组:正常对照组(NC组)、糖尿病组(DM组)、EGB治疗组(EGM组,8mg.kg-1.d-1)。腹腔注射链脲佐菌素(STZ,30mg/kg)诱导大鼠造成糖尿病模型,EGB组用银杏叶提取物进行腹腔注射8周后,用免疫组化法检测各组大鼠肾皮质MMP-2、MMP-9的蛋白表达。结果:免疫组织化学:DM组大鼠肾小球MMP-2和MMP-9蛋白表达较正常对照组明显减少(P<0.01);EGB组MMP-2和MMP-9较DM组表达升高(P<0.01)。结论:EGB可通过上调MMP-2和MMP-9蛋白表达,减少细胞外基质积聚而对糖尿病大鼠肾组织损伤起到一定的治疗作用。
Objective:To study the expressions of matrix etalloproteinase-2(MMP-2),matrix metalloproteinase-9(MMP-9) on the kidney tissue of diabetic rats after the treatment of EGB.Method: 50 adult SD rats were randomly divided into three groups,normal control group(NC group),diabetes mellitus group(DM group) and diabetic group treated with extract Gingko biloba(8mg.kg-1.d-1)(EGB group).Diabetes model was induce by streptozotocin(STZ,30mg/kg) and remedial injected by administrating EGB was done on the EGB group 8 weeks later,the morphological changes were observed though the light microscope and the expression of MMP-2,MMP-9 in kidney tissue was determined by immunohistochemistry methods.Results: Immunohistochemical analysis showed that compared with the DC group,the protein expression of MMP-2 and MMP-9 were significantly decreased in DM group,and compared with the DM group,the protein expressions were significantly increased in EGB group.Conclusion: EGB had protective effects on kidney tissue of diabetic rats through increasing the expression of MMP-2 and MMP-9.
