摘要
目的:探讨硫化氢对小鼠3T3-L1前脂肪细胞增殖的影响。方法:培养3T3-L1前脂肪细胞,分为对照组、低剂量组和高剂量组(硫化氢浓度分别为0、10、25、50和100μmol/L),采用四甲基偶氮唑盐比色(MTT)法检测硫化氢对3T3-L1前脂肪细胞活性的影响;用BRDU法检测细胞的增殖;结果:与对照组相比,H2S在较高浓度时,对细胞有一定的毒性作用(P<0.05)。结论:硫化氢抑制3T3-L1前脂肪细胞的增殖。
Objective:To disscuss the influence of hydrogen sulfide which actson 3T3-L1 preadipocyte of mouse.Methods:Cul tured the 3T3-L1 preadipocyte,and then divided into three groups:the control group,the low monitoring group and the high monitoring group(hydrogen sulfide density(0μmol/L,10μmol/L,25μmol/L,50μmol/L and 100μmol/L,respectively) using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) colorimetric.Method:to measure the viability of 3T3-L1 preadipocyte to which added hydrogen sulfide.Cell proliferation rate was measured by BrdU.Results: H2S has certain toxic effect on the cell at high concentration compared with the control group(P 0.05).Conclusion:The hydrogen sulfide inhibits 3T3-L1 preadipocyte's proliferation.
