内脂素对人胆管癌细胞QBC939侵袭及增殖影响的体外研究

摘要 目的:研究人内脂素(visfatin)对胆管癌细胞QBC939体外增殖和侵袭能力的影响,并初步探讨可能存在的机制。方法:培养胆管癌细胞QBC939,在不同浓度的内脂素作用不同时间后,以四甲基偶氮唑盐(MTT)法检测细胞增殖能力,用Tran-swell小室检测内脂素(200 ng/ml)24 h后对胆管癌QBC939细胞侵袭能力的影响,Western blot检测内脂素对QBC939细胞内基质金属蛋白酶(matrix metalloproteinase,MMP)-2蛋白的表达。结果:内脂素可以促进QBC939细胞的增殖(P<0.01),且存在剂量依赖性;内脂素(200 ng/ml)处理24 h后,QBC939细胞穿透聚碳酸酯膜的能力与对照组相比显著增强(P<0.01);Westernblot显示内脂素(200 ng/m1)处理24 h后,QBC939细胞MMP-2蛋白的表达明显增高。结论:内脂素可以明显提高人胆管癌细胞QBC939的增殖和侵袭能力,其作用机制可能与MMP-2水平的增高有关。 Objective:To investigate the effects of visfatin on the proliferation and invasion of human cholangiocarcinoma QBC939 cells,and explore the possible mechanisms.Methods:QBC939 cells were cultured and treated with visfatin at different concentra-tions of 25,50,100 and 200 ng /ml for 6,12,and 24 h.QBC939 cells without visfatin treatment were used as control group.The pro-liferation rate of the QBC939 cells was measured by MTT assay.Transwell chamber assay was performed to determine the effect of visfatin 200 ng /ml on the invasion capability of QBC939 cells for 24 h later,and Western blot was used to observe MMP-2 protein expression.Results:The results showed that visfatin could significantly improve growth and proliferation of QBC939 cells.The effect is in a dose and time-depended by MTT method(P 〈 0.01).The invasion capability of QBC939 cell was significantly increased after visfatin 200 ng /ml treatment for 24 h(P 〈 0.01).The expression of MMP-2 was increased in QBC939 cells treated with visfatin.Conclusion:Visfatin significantly increases the proliferation and invasion capabilities of QBC939 cells in vitro.The possible mecha-nism may be related with up regulation of MMP-2 level.
