摘要
目的建立BGC-823胃癌细胞株的裸鼠成瘤模型,为胃癌研究提供参考载体。方法体外培养BGC-823细胞,常规传代,收集并用生理盐水冲洗细胞,并用生理盐水制成细胞密度为22×1010/mL的单细胞悬液,取0.1 mL注入裸鼠颈部与前肢交界处皮下。观察并测量裸鼠成瘤情况,切片光镜观察瘤体组织结构。结果裸鼠注射后未发生明显的炎症反应,1周时虽肉眼观察不到瘤体,但能触及实体豆状颗粒;2周时即能观察到瘤体形成,触诊能扪及实体包块;4周时瘤体明显,呈分页状,直径可达0.9cm;8周时瘤体巨大,凹凸不平,直径可达3.0 cm以上。光镜下可见瘤体组织癌细胞核大,多呈椭圆形或不规则形,排列杂乱。结论经皮下注射BGC-823细胞能有效建立裸鼠胃癌成瘤模型。
Objective To build gastric carcinoma oncogenesis model in nude mice with BGC-823 cell strain and to provide references to related researches.Methods BGC-823 cell strain was cultured in vitro and passaged by conventional methods.The cells were collected and washed by normal saline,which was later made into single-cell suspension with cell density of 22×1010/mL.0.1 mL single-cell suspension was injected subcutaneously into the cross point of neck and fore limb in nude mouse.The oncogenesis was observed and measured,meanwhile the organization structure was observed through slices under light microscope.Results There was no symptom of infection in nude mice after the injection.The tumor can not be seen by naked eyes at 1st week but solid bean-like pallet can be felt.The oncogenesis can be observed at 2nd week and solid mass can be felt by palpation.At 4th week,the tumor became obvious;it was page-like with diameter of 0.9 cm.At 8th week,the surface of the tumor became uneven and the tumor was large in volume with diameter of 3.0 cm above.The cell nucleus was big,oval or irregular in shape,organized in chaos under light microscope.Conclusion Subcutaneous injection of BGC-823 cell can effectively build gastric carcinoma oncogenesis model in nude mouse.
