摘要 目的:探讨急性冠脉综合征患者Th17细胞和Treg细胞的变化及意义。方法:选取51例急性冠脉综合征患者,25例稳定性心绞痛(SA)患者,27例同期住院的健康对照者,分别采用流式细胞术、实时定量PCR和ELISA法检测外周血Th17细胞和Treg细胞百分率、关键核转录因子RORγt、STAT3和Foxp3 mRNA的表达以及血浆IL-17A和TGF-β1的浓度。结果:①ACS患者外周血中Th17细胞百分率、RORγt和STAT3 mRNA显著高于SA患者和对照组(P<0.01);②与SA组和对照组相比,ACS患者CD4+CD25+Foxp3+Treg和CD4+Foxp3+Treg细胞百分率,Foxp3 mRNA的表达以及血浆TGF-β1的浓度显著降低(P<0.01)。结论:ACS患者外周血中Th17/Treg失衡,Th17/Treg失衡可能与斑块的不稳定密切相关。 Objeclive:To study the changes and significance of Tbl7 cell and regulatory T cell in patients with acute coronary syndrome(ACS). Methods:51 patients with acute coronary syndrome, 25 patients with stable angina (SA) and 27 healthy control subjects were consecutively enrolled. The percentage of Thl7 cells and Treg cells, mRNA expression of key transcription factors (RORyt, STAT3, Foxp3) and plasma IL-17A in these patients were measured by flow cytometry, real-time quantitative PCR and ELISA. Results: (DThe percentage of Thl7 cells, mRNA expression of RORTt and STAT3 increased significantly in ACS group compared with SA group and controls( P 〈 0.01 ) ; ②The percentage of CD4^+ CD25^+Foxp^3+ Treg cells and CIM^+ Foxp3^+ Treg cells, mRNA expression of Foxp3 and plasma TGF-β1 were significantly lower in ACS group than in SA group and controls (P 〈 0.01 ), while there was no significant difference in percentage of CIM ^+CD25^+ Treg cell among 3 groups. Conclusion:The imbalance of Thl7/Treg exists in ACS patients, which may contribute to plaque destabilization.
