摘要
目的建立树突膜片钳技术,观察全细胞记录模式下大鼠视皮层脑片锥体神经元树突部位电生理性质。方法 采用红外微分干涉相差显微镜结合电耦合式摄像机,利用膜片钳技术进行近端树突钳制,成功形成全细胞记录模式后将膜电位钳制在不同水平,记录大鼠视皮层锥体神经元树突部位的电生理性质。结果 成功记录到电流钳模式下树突部位膜电位,及膜电位钳制在不同水平时电压钳下记录到的突触后自发及诱发反应。结论 采用可视法树突膜片钳技术可以记录到树突部位膜电位、自发及诱发反应,这项新技术对树突部位电生理性质及其上分布的离子通道功能的深入研究具有重要意义。
Objective To investigate membrane electrophysiological properties of dendrites by whole-cell recording with dendritic patch-clamp technique. Methods Experimental study was taken. By combining infrared differential interference contrast (IR-DIC) technique and CCD-Camera system, clamped proximal dendrites with patchclamp technique, when the whole-cell recording mode was successful formed, recorded the electrophysiological properties of pyramidal neurons dendrites in rat visual cortex with different holding potentials. Results Successful records were taken at mem- brane potential of dendritic with the current clamp mode, and spontaneous and evoked postsynaptic responses with voltage-clamp mode when membrane potential was held in different levels. Conclusions Visualized dendritic patch-clamp recording technique can be used to record the membrane potential of dendrites with the current clamp mode, and the spontaneous and evoked postsynaptic responses with the whole-cell recording of dendritic patch-clamp technique, and this new technology is important for further study electrophysiological properties and ion channels of dendrites.
