黄芪对卵巢切除术后大鼠皮肤生物酶及形态的影响

摘要 目的建立绝卵巢切除术后皮肤老化模型,观测黄芪提取液对卵巢切除术后大鼠皮肤衰老的影响。方法将60只Wister雌性大鼠随机分成4组:非手术组、手术组(切除双侧卵巢正常喂养组)、雌激素给药组和黄芪给药组。正常饮食、饮水,饲育50d后处死大鼠,取皮肤组织测定其生物活性酶及病理组织观察。结果与非手术组比较,手术组皮肤组织中SOD活性明显降低,羟脯氨酸含量显著减少,丙二醛含量显著增高(P<0.05)。雌激素组及黄芪组皮肤组织中SOD活性不降低,羟脯氨酸含量无减少,丙二醛含量不增高(P>0.05)。与手术组比较,黄芪组皮肤组织中SOD活性明显增高,羟脯氨酸含量增加,丙二醛含量降低(P<0.05),与雌激素组无显著性差异(P>0.05)。组织病理示:,卵巢切除术后皮肤组织老化,雌激素和黄芪治疗后可改善和延缓其皮肤衰老。结论切除大鼠双侧卵巢可以建立绝经期皮肤老化模型,黄芪提取液对大鼠皮肤衰老具有延缓作用,与激素替代疗法无显著性差异。 Objective To establish the menopause mice model by ovariectomized method. To observe the influence of astragalus anti-skin aging to menopause mice. Methods Sixty wister male mice were randomly divided into four groups, including nomal control group and other 3 groups. To make animal models, Wister male mice were ovariectomy control, the experimental groups included operation model group( without further treatment group), astragalus treatment group(8g/kg)and hormone replacement therapy group( injection estradiol benzo- ate 200ug/kg). The dorsal skin samples were collected to determine biochemical analyses were carried out at 50 days after the operation. And observed dorsal skins under the microscope and analyzed. Results Com- pared with the nomal control group, the content of MDA in the ovariectomy group had significantly increased (P 〈 0.05 ). The astragalus treatment group and hormone replacement therapy group had no change ( P 〉 0.05 ). However, the activity of SOD and HYP content had statistical significance and to be significantly de- creased(P 〈 0.05). The astragalus treatment group and hormone replacement therapy group had no change compared with the normal group(P 〉 0.05 ). In contrast with the non-operation group, the operation group showed that the thickness of dermis was significantly reduceed and the content of elastic fibers was de- creased. Conclusion The astragalus treatment group and hormone replacement therapy group had no change. From the exepriment statistical, it was found that: Astragalus can clean free radicals and has antioxi- dantive function to menopause mice for anti-aging of the skin.
