摘要
目的结合形态学技术分析免疫缺陷小鼠的面神经损伤特点,为进一步深入探讨面神经修复再生的神经免疫病理机制提供实验基础。方法制备周围性面神经外伤模型。分别于术后3 d、2周灌注固定动物,收集脑干切片,免疫组化分析模型小鼠面运动神经元诱导型一氧化氮合酶(inducible nitricoxide synthase,iNOS)表达情况。并结合面神经周围主干锇酸染色情况分析裸鼠面神经损伤特点。结果免疫组化显示裸鼠术后3 d受损伤的面运动神经元iNOS强阳性表达,间以无iNOS表达的"健康"面运动神经元;裸鼠术后14 d术侧面运动神经元的诱导型iNOS已基本无表达。结论面神经轴切损伤早期,损伤的面运动神经元有诱导型iNOS表达,并存在一个时间动力学改变,这可能与面神经损害的发生及神经修复的启动相关。
OBJECTIVE To analyze the characteristic of facial nerve injury on immune deficiency mouse model with facial nerve axotomy by morphological technique in order to reveal the latent neuroimmunologic pathophysiological mechanism of traumatic facial paralysis.METHODS The nude mouse(BALB/c background)were subjected to a right facial nerve axotomy.The expression of inducible nitricoxide synthase in facial motoneuron were analyzed after collecting the slices of brain stem at specific time.The characteristic of facial nerve injury were assessed with osmic acid staining.RESULTS There was strong expression of inducible nitricoxide synthase in facial motoneuron of nude mouse with axotomy at 3 days postoperatively;whereas no expression of inducible nitricoxide synthase was found at 14 days postoperatively.CONCLUSION There was expression of inducible nitricoxide synthase in facial motoneuron of nude mouse with facial nerve axotomy at early stage.And the expression was in a way of time-dependant alteration.It may relevant to the onset of facial nerve injury and the start of nerve repair.
