宫颈癌脱落细胞人端粒酶逆转录酶mRNA表达临床意义的探讨 被引量:4

Expression of hTERT mRNA in exfoliated cells of cervical cancer and its clinical significance

摘要 目的:探讨人端粒酶逆转录酶(hTERT)mRNA在宫颈癌(CC)脱落细胞中的表达及其与临床病理参数之间的关系。方法:采用实时荧光定量RT—PCR技术,检测人宫颈癌HeLa细胞株(细胞组)、40例宫颈癌(宫颈癌组)和21例正常或慢性宫颈炎(对照组)患者宫颈脱落细胞中hTERT mRNA的表达,以细胞组hTERT mRNA的表达作为定性、定量参照。结果:宫颈癌组和对照组中hTERT mRNA阳性表达率分别为82.5%和9.5%,差异有统计学意义,P=0.000;两组hTERT mRNA的相对定量水平NhTERT分别为0.3276~13.7823和0.0093~0.0816,差异均有统计学意义,P=0.019;hTERTmRNA阳性表达与宫颈癌的分化程度具有负相关性(r=-0.308,P=0.016),与年龄、临床分期、间质浸润深度和组织学分类无相关性。结论:宫颈癌患者宫颈脱落细胞hTERT mRNA阳性表达率及相对定量水平均异常增高,检测宫颈脱落细胞hTERT mRNA的表达有助于宫颈癌的诊断及预后评估。 OBJECTIVE: To study human telomerase reverse transcriptase(hTERT) mRNA in cervical exfoliated cells from the specimens of normal and cervical cancer(CC), and analyze its relation with clinical or patho-characters and its contribution to clinical diagnosis. METHODS: The real-time fluorescent quantitative reverse transcription polymerase chain reaction(real-time qRT-PCR) was employed to detect the hTERT mRNA positive expression and quantitative levels of HeLa cell strains(the cell group),and of cervical exfoliated cells in 40 cases of CC(the cancer group)and 21 cases of normal or chronic cervicitis(the control group). RESULTS: The positive rate of hTERT mRNA in exfoliated cells from the cancer group and the control group were expressed as 82. 5% and 9.5%(P=0. 000). The quantitative levels of hTERT mRNA(NhTERT) in them were respectively expressed as 0. 327 6 - 13. 782 3 and 0. 009 3-0. 081 6(P=0. 019). All of them indicated statistical significance. In the cancer group, there is relativity between the positive rate of hTERTmRNA and differentiation of CC (r= -0. 308, P = 0.016), but there is no relativity among age, clinical stages, invasive depth and histological classification. CONCLUSIONS: The positive rate and quantitative levels of cervical exfoliated cells from specimens of CC are more higher than that of the normal cervix. The proceture is recommended for early detection in clinic for diagnosing CC and its prognosis.
