高压氧下大鼠脑缺血区域脑组织MMP-9表达及作用 被引量:2

摘要 目的研究高压氧下大鼠脑缺血区域脑组织基质金属酶-9(MMP-9)表达及促进大鼠脑缺血损伤恢复的机制。方法将32只大鼠分为模型组(16只)和假手术组(16只),两组均分别行高压氧治疗(8只)和高压空气治疗(8只)。高压空气组与高压氧治疗组在时间、治疗次数方案上相同,均1次/d,共3d。大鼠治疗第4天分别用半湿法检测脑含水量,脑组织伊文思蓝(EB)定量检测脑组织,ELISA试剂盒检测MMP-9。结果治疗第4天,大鼠模型组在高压氧治疗下脑含水量、脑组织EB定量及MMP-9含量均小于高压空气下的治疗(均<0.05)。假手术组用高压氧治疗和高压空气治疗差异均无统计学意义(均>0.05)。结论 MMP-9参与脑缺血再灌注损伤后的炎症反应过程,高压氧治疗下可通过抑制MMP-9表达,干扰脑缺血再灌注损伤后炎症的发生发展过程,减轻血脑屏障的破坏,继而减轻脑水肿,发挥良好的脑保护作用。 Objective To investigate the effect of hyperbaric oxygen(HBO)onactue focus and expression of MMP-9 in the cerebral ischemia of rats.Methods 32 Wistar rats were randomly divided into two groups,model group(=16) and sham-operation group(=l6).In both group,8 rats were treated with HBO treatment and the other ones with high pressure air.The time of each therapy and treating times were the same in both groups,once a day,for 3 days.On the 4th day,cerebral edema formation was determined by brain water content;the histological morphological damage was investigated by EB staining;MMP-9 was observed by ELISA reagents.Results On the 4th day,among model gro-up,moisture content of brain,EB content of brain tissue and MMP-9 content of rats with HBO treatment were less than those of rats with air treatment,with significant differences( 0.05);but there were no significant differences among shan-operation group.Conclusions MMP-9 might contribute to inflammatory reaction after ischemia reper-fusion cerebral.HBO treatment can disturb the development of inflammatory reaction,and further play a neuro pro-tective role in ischemia cerebral.
