山药多糖对CCl_4肝损伤小鼠自由基、TNF-α含量的影响 被引量:13

摘要 目的探讨山药多糖对CCl4肝损伤小鼠NO、TNF-α含量的影响。方法昆明种小鼠50只随机分正常对照组,病理模型组,山药多糖高、中、低浓度组。山药多糖高、中、低浓度组每天按体重30 mg/kg,60 mg/kg,120 mg/kg山药多糖灌胃,饲养8 d,末次灌胃2 h,对照组腹腔注射调和油溶液,其余各组腹腔注射0.15%CCl4调和油溶液,24 h眼球取血分离血清,测定ALT活性。处死小鼠取出肝脏称重计算肝体指数,制备肝匀浆,按试剂盒要求测定MDA、NOS、NO、TNF-α的含量。结果山药多糖可减轻实验性肝损伤所致炎性反应,降低肝体指数,降低血清中ALT活性(P<0.05);降低肝脏MDA、NOS、NO和TNF-α的含量(P<0.01)。结论山药多糖可通过降低MDA、NO和TNF-α的含量对CCl4肝损伤有保护作用。 Objective To explore the effect of yam polysaccharide on the contents of free radicals and TNF-α in mice of CCl4-induced liver injury. Methods Fifty Kun-ming mice were randomly divided into control group,model group and low,medium,high dosage of yam polysaccharide groups.The mice in yam polysaccharide groups were given 30 mg/kg,60 mg/kg,120 mg/kg of yam for 8 d,respectively.The mice were intraperitoneally injected with blend oil in control group at 2 h after the last administration,and the mice in other groups were intraperitoneally injected with 0.15% CCl4 mixed with blend oil.The activity of ALT in blood serum was determined after 24 h.The liver index was calculated and the contents of MDA,NOS,NO and TNF-α in liver homogenate were determined. Results Yam polysaccharide alleviated the inflammation in response to experimental liver injury,decreased the liver index and ALT activity(P〈0.05),and it also decreased the contents of MDA,NOS,NO and TNF-α in the liver(P〈0.01). Conclusion Yam polysaccharides can protect the CCl4-induced liver injury by reducing the contents of MDA,NO and TNF-α.
