摘要
由动物标本提取动物DNA,对于识别动物的物种、了解动物的变异以及遗传多样性具有重要意义.作者对20世纪50年代左右的林麝熟制标本皮张进行了DNA提取,并用林麝细胞色素b基因引物进行PCR扩增,对扩增的结果进行了测序,结果表明标本的核DNA含量少,线粒体DNA保存较好,能够特异性扩增出,与新鲜肉提取的DNA相同.
DNA extraction from animal specimens is significant for identification of animal species and investigation on variation and genetic diversity of animals. This paper exact DNA from dressed skin in Moschus berezovski flerov specimen made 50% last century. Cytochrome gene b was utilized in process of primer to amplify PCR. The result was sequenced and showed that specimen was of lower nuclear DNA content, better conserved mitochondrial DNA, and it could be amplified as DNA extracted from fresh flesh.
