摘要
目的:初步探讨桩蛋白(paxillin)在大鼠星形胶质细胞及神经元中的表达。方法:通过组织切片、细胞培养、免疫荧光和RT-PCR方法,观察桩蛋白在大脑皮层及体外培养的星形胶质细胞以及神经元中的表达。结果:桩蛋白在大鼠脑皮质的星形胶质细胞中表达,神经元未见明显表达;在体外培养的星形胶质细胞以及神经元中的表达情况与体内一致,主要表达于星形胶质细胞的胞浆及突起中。结论:桩蛋白主要表达于星形胶质细胞而非神经元中。本实验结果为进一步研究桩蛋白在星形胶质细胞中的生物学作用奠定了基础。
Objective: To study the expression of paxillin in astrocytes and neurons of rat brain.Methods: The expression of paxillin was investigated in astrocytes and neurons by tissue section,cell culture,immunofluorescence and RT-PCR methods.Results: In the cortex,paxillin expressed mainly in astrocytes instead of the neurons.RT-PCR showed that paxillin mRNA was present in cultured astrocytes,but not in cultured neurons.Immunocytochemistry revealed that paxillin was located in the astrocytic cytoplasm and cell processes but not in the nucleus.Conclusion: Paxillin was expressed in astrocytes not in neurons.These findings contribute to a clearer understanding of paxillin in astrocytes.
