摘要
目的:观察大鼠创伤性脑损伤(TBI)后不同时相点损伤区周围皮质胰岛素样生长因子-1(IGF-1)mRNA和蛋白的表达变化。方法:采用大鼠自由落体脑损伤模型(Feneey法),通过RT-PCR和Western Blot方法检测大鼠TBI后12、24、48、72 h损伤区周围皮质IGF-1 mRNA和蛋白的表达,用免疫荧光检测上述相同时间点IGF-1的表达定位。结果:大鼠TBI后12 h损伤区周围皮质IGF-1 mRNA和蛋白已开始显著表达,24 h达到高峰,48 h表达开始减少,72 h仍有少量表达。结论:大鼠TBI后,损伤区周围皮质IGF-1的表达在伤后24 h达到高峰,48 h后开始减少的变化趋势可为应用IGF-1治疗TBI提供合适的时间窗。
Objective: To observe the time-course expression of insulin-like growth factor-1(IGF-1)in cerebral cortex lesion of rats with traumatic brain injury(TBI).Methods: Sprague-Dawley(SD) rat models with traumatic brain injury were made by Feeney's method.Then reverse transcriptase-polymerase chain reaction(RT-PCR) analysis,Western Blot were used to detect the expression and location of IGF-1 in cerebral cortex lesion of rats with TBI at 12,24,48,72 h after TBI.Results: The expression level of IGF-1 started to increase from 12 h after TBI.The peak of expression appeared at 24 h after TBI.The expression decreased from 48 h and a small amount of expression were observed at 72 h after TBI.Conclusion: The tendency of endogenic IGF-1 expression after TBI might provide the proper time-window of treatment to TBI.
