摘要
目的观察姜黄素对血管紧张素Ⅱ(AngⅡ)诱导的乳鼠原代培养心肌细胞血凝素样氧化低密度脂蛋白受体-1(LOX-1)mRNA表达的影响。方法体外培养SD大鼠乳鼠原代心肌细胞,按细胞处理方式不同分为4组:阴性对照组(A组),AngⅡ组(B组),AngⅡ+LOX-1抗体组(C组),AngⅡ+姜黄素组(D组)。C、D组分别给与LOX-1抗体(10μg/ml)、姜黄素(5μmol/L)预处理3 h,后给与AngⅡ作用24 h。RT-PCR法检测细胞LOX-1 mRNA的表达。结果 B组LOX-1 mRNA表达较A组明显上调(P<0.05),C、D组LOX-1 mRNA表达较B组明显下调(P<0.05),C组与D组比较,差异无统计学意义。结论姜黄素可以下调AngⅡ诱导的乳鼠原代心肌细胞LOX-1 mRNA的表达。
Objective To investigate the effects of curcumin on the mRNA expression of lectin-like oxidized low density lipoprotein receptor 1(LOX-1) in cultured cardiomyocytes by angiotensin Ⅱ(AngⅡ).Methods The cultured cardiomyocytes were divided into four groups:the negative control group(group A),treated with AngⅡ(group B),the AngⅡ+anti-LOX-1 antibody group(group C),the AngⅡ+curcumin group(group D).Group C and group D were separately treated with anti-LOX-1 antibody(10 μg/ml)and curcumin(5μmol/L)for 3 h,and then stimulated with AngⅡ for 24 h.LOX-1 mRNA expression was detected by RT-PCR.Results LOX-1 mRNA expression in group B was obviously upregulated than that in group A(P0.05).Pretreatment of cells with curcumin reduced the increase of LOX-1 mRNA expression which treated with Ang Ⅱ(P0.05).The expression of LOX-1 mRNA was not significantly different compared with pretreatment with anti-LOX-1 antibody(P0.05).Conclusion Curcumin may down-regulate the expression of LOX-1 mRNA which may be induced by AngⅡ.
