口腔鳞癌组织中PTEN、COX-2表达和MVD测定及意义 被引量:3

Expression and significance of PTEN,COX-2 and MVD in OSCC

摘要 目的研究第10号染色体缺失与张力蛋白同源的磷酸酶(PTEN)、环氧化酶-2(COX-2)在口腔鳞癌(OSCC)组织中的表达及其与临床病理特征和肿瘤血管生成的关系。方法采用免疫组化SP法分别检测50例OSCC组织和12例正常口腔黏膜组织中PTEN、COX-2、CD34的表达。结果 PTEN和COX-2阳性率在正常口腔黏膜为100%和0,OSCC组织中为56.0%与58.0%,微血管密度(MVD)为(45.63±10.52)。PTEN阴性表达与OSCC的分化程度和临床分期相关(P<0.05),COX-2阳性表达与临床分期、分化程度、淋巴结转移相关(P<0.05);PTEN阳性表达组织中的MVD值为(41.17±10.46),阴性组织为(47.46±12.31),两者差异有统计学意义(P<0.01)。COX-2阳性表达组织中的MVD值为(45.32±12.87),阴性组织为(37.18±9.68),两者差异有统计学意义(P<0.01)。PTEN和COX-2表达呈显著负相关(rs=-0.509,P<0.01)。结论 PTEN失活、COX-2的激活及血管的形成在OSCC进程中共同发挥作用。 Objective To investigate expression of phosphatase and tensin homologue deleted on chromosome ten(PTEN) and cyclooxygenase-2(COX-2),and their relationship with angiogenesis in oral squamous cell carcinoma(OSCC).Methods PTEN,COX-2,CD34 were detected by immuohistochemical SABC method in 50 cases of OSCC,12 cases nomal oral mucosa.Results The positive rates of PTEN and COX-2 expression were 100% and 0.0% in nomal mucosa,56.0% and 58.0% in OSCC tissue.Microvascular density(MVD) was(45.63±10.52).The PTEN expression was significantly associated with differentiation and clinical stages(P0.05).The COX-2 expression was significantly associated with differentiation,clinical stages and lymph node metastasis status(P0.05).In the PTEN positive expression cases of MVD were(41.17±10.46),in the PTEN negative expression cases of MVD were(47.46±12.31),there was significant difference in two groups(P0.01).In the COX-2 positive expression cases of MVD were(45.32±12.87),in the COX-2 negative expression cases of MVD were(37.18±9.68),there was significant difference in two groups(P0.01).The expression of PTEN was negative correlated with COX-2(rs=-0.509,P0.01).Conclusion PTEN inactivation and COX-2 overexpression with angiogenesis play an important role in tumor progression in OSCC.
