聚二磷酸腺苷核糖聚合酶-1在细胞死亡中的作用

摘要
聚二磷酸腺苷核糖化是细胞应对DNA损伤、保持基因组完整的一种重要机制。负责催化该反应的酶-聚二磷酸腺苷核糖聚合酶-1(PARP-1)参与了DNA损伤导致的两种细胞死亡:细胞凋亡和细胞坏死。细胞凋亡时,PARP-1被caspase特异性地降解,产生氨基端的24 ku片段和羧基端的89 ku片段;而完整的PARP-1的过度活化会引起能量丧失而致细胞坏死。现对PARP-1在这两种死亡结局中的作用作一综述。
