孕鼠叶酸缺乏对幼鼠肺发育及叶酸结合蛋白1基因的影响 被引量:1

Effects of folic acid deficiency during pregnancy on pulmonary development and expression of foliate binding proteins gene one in neonatal rats

摘要 目的探讨孕鼠叶酸缺乏对幼鼠肺发育及其叶酸结合蛋白1基因(Folbp1)的影响。方法成熟雌性36只SD大鼠随机均分为A组(缺乏叶酸饲料的实验组)和B组(添加叶酸饲料的对照组),2周后与成熟SD雄性大鼠交配,生后7 d处死新生大鼠,留取肺组织标本。HE染色观察病理改变,实时荧光定量RT-PCR技术检测Folbp1 mRNA表达。结果与B组比较,A组肺组织结构紊乱,肺泡数量明显减少,肺间隔明显增宽,肺泡、孔腔形状较不规则,肺组织结构明显被破坏。A组肺组织中Folbp1 mRNA相对表达量低于B组(561.21±73.53 vs.1095.73±95.88)(P<0.05)。结论孕期叶酸缺乏会降低Folbp1基因表达,从而可能导致幼鼠肺发育形态异常,造成肺发育成熟障碍。 Objective To investigate the effects of folic acid deficiency during pregnancy on pulmonary development and expression of folic acid binding proteins gene 1(Folbp1) in neonatal rats.Methods Thirty-six adult female SD rats were randomly and equally divided into two groups of A(fed without folic acid) and B(fed with folic acid).After 2 weeks,the female rats in both groups were copulated with male SD rats.The neonatal rats were sacrificed on the 7th day after birth,and the lung samples were collected.The changes of pathobiology were detected by HE staining,and the expression of Folbp1 mRNA was determined by real-time fluorescence quantitative-PCR.Results Compared with group B,the structural disorder of lung tissue,decreased number of lung alveolus,enlarged lung interval,irregular shape of alveolar and cavity holes and destroyed structure of lung tissue were observed in group A.The expression of Folbp1 mRNA in group A was lower than that in group B(561.21±73.53 vs.1095.73±95.88)(P〈0.05).Conclusion Folic acid deficiency during pregnancy can decrease the expression of Folbp1,which may lead to the abnormal morphology of lung development and the barrier of lung maturation.
