实验性脂肪肝大鼠胃窦Ghrelin的研究

摘要 目的建立脂肪肝大鼠模型,观察脂肪肝胃肠动力紊乱模型中大鼠胃窦组织内Ghrelin的表达,探讨Ghrelin与脂肪肝胃肠功能紊乱的关系,为临床胃肠动力异常疾病的治疗提供理论依据。方法 SD大鼠20只,随机分为正常对照组(n=10)和脂肪肝组(n=10)。采用高脂饮食制备大鼠脂肪肝模型,3个月后用放射性核素99mTc灌胃测定大鼠胃液体排空率,取正常对照组及模型组大鼠胃窦标本,S-P免疫组化染色法对Ghrelin免疫反应阳性结构进行染色及分析。结果脂肪肝模型复制成功。与正常大鼠相比,胃排空率脂肪肝组(86.14±2.40)%较对照组(89.45±1.85)%减弱(P<0.01),胃窦Ghrelin阳性单位数脂肪肝组(32.59±15.78)较对照组(51.89±10.65)明显降低(P<0.01)。结论脂肪肝大鼠存在胃运动功能减退;脂肪肝大鼠胃运动功能减退与Ghrelin的减少存在一定相关性。 Objective To observe the expression of Ghrelin in gastric antrum of rats with fatty liver and to explore the relationship between gastric motility of fatty liver and Ghrelin, then provide a theoretical basis for the clinical treatment of gastrointestinal disorders. Methods Twenty SD rats were selected and randomly divided into fatty liver group (n = 10) and normal control group (n= 10). Fatty liver model were duplicated by high fat diet feeding. Using gastrogavage with radioactive 99mWc to quantify liquid food emptying rate after three months. The tissue specimens were obtained from both groups. The expression of Ghreliwpositive unit was analyzed by S-P immuonhistochemistry. The results were investigated. Results Duplicating successful models of fatty liver. Compared with that of the controls, the gastric emptying of rats with fatty liver decreased(86.14 ± 2.40)% vs (89.45± 1.85) % (P〈0.01) ,the positive units of Ghrelin decreased(32.59± 15.78)vs(51.89 ± 10.65) (P〈0.01). Conclusion The gastric motility of rats with fatty liver was markedly delayed, which may be associated with Ghrelin.
