摘要 细胞毒物、致癌物以及亲电子试剂在外界环境中无处不在,这些物质及其代谢产物直接或间接干扰DNA、蛋白质和脂质等生物大分子的生理功能,参与各种疾病包括肿瘤、老化、哮喘、急性肺损伤、 Nuclear factor-E2-related factor 2(Nrf2) plays a critical role in the regulation of cellular redox status.Under normal homeostatic conditions,Nrf2 transcription is repressed by its negative regulator Kelch-like epichlorohydrin-associated protein 1(Keap1).However,upon exposure to oxidative stress,Nrf2 dissociates from cytosolic Keap1 and translocates to the nucleus,where it binds to the corresponding response element(ARE) in the promoter region of the genes encoding antioxidant enzymes and phase 2 detoxifying enzymes to induce the expression of endogenous corresponding enzymes.Nrf2-induced antioxidant protection involves in cancer,inflammation,respiratory diseases,cardiovascular diseases,neurodegeneration,autoimmune diseases and so on.On the contrary,disruption or inactivation of Nrf2 will aggravate the cytotoxicity of oxidative stress,leading to the cellular dysfunction,apoptosis or even death.The purpose of this review is to introduce the interaction between Nrf2 and Keap 1,and the latest research progress on the function of Nrf2.
