湿地植物水芹再生体系的建立与优化 被引量:1

Establishment and optimization of high-frequency regeneration system for Oenanthe javanica

摘要 以水芹种子胚为外植体,通过离体培养技术,建立高频的水芹再生体系。以MS为基本培养基,添加不同种类和不同浓度的植物激素,探讨影响水芹离体组织培养的系统因子。结果表明:愈伤组织诱导和继代培养基较适宜比为:MS+0.5 mg.L-16—BA+1.0 mg.L-1NAA+2,4—D 0.1 mg.L-1,诱导率达67.5%;较适宜分化诱导培养基为:MS+0.2 mg.L-16—BA+0.05 mg.L-1NAA,继代天数为42 d;较适宜生根培养基为:1/3 MS+IAA0.6 mg.L-1+NAA 0.05 mg.L-1,生根率为89.5%;移栽至炉灰渣+蛭石+细沙=1∶1∶1或者珍珠岩+蛭石=2∶3的人工土壤中,成活率达95%,人工苗长势良好。 A high-frequency regeneration system of Oenanthe jawanica has been established by means of in vitro culture technology with the seeds of O. javanica as explants. By taking MS media added different hormones with various concentrations, vitro tissue culture of O. javanica was carried out to study the effects of the conditions to the system. The results show that the optimal medium for callus induction and secondary culture was MS+0.5 mg · L^-16-BA+ 1.0 mg · L^-1 NAA+ 0. 1 mg · L^-12,4-D, the callus induction rate reached 67. 5%; the best medium for callus differentiation was MS+0. 2mg · L^-16 BA4- 0.05 mg · L^-1NAA, the callus were developed by 42 days every time; the best medium for rooting induction was 1/3MS + 0. 6 mg· L^-11AA+0. 05 mg· ^-1 NAA, the rooting rate was 89. 5%; when they were transplanted to the medium composed with one part furnace ash, one part vermiculite and one part silver sand or two parts perlite and three parts vermiculite, the survival rate was 95% and the artificially seedlings grew well.
