摘要
水稻OsMADS3基因编码一个MADS盒转录因子,在早期花器官决定过程中发挥着重要作用。为进一步研究OsMADS3的生物学功能,构建了pET32a-OsMADS3原核表达载体,在大肠杆菌BL_(21)(DE_3)中成功表达了带有6×His标签的OsMADS3全长重组蛋白,占细胞总蛋白的60%,主要以包涵体形式存在,分子量为48kD,与预期大小一致。
The rice OsMADS3 gene encodes a MADS-box transcription factor,which plays an important role in the early floral organ-determining process.To further reveal the biological function of OsMADS3,a prokaryotic expression vector of pET32a-OsMADS3 was constructed,and the OsMADS3 full length recombinant protein with a 6xHis label was successfully expressed in Escherichia coli strain BL_(21)(DE_3).The fused protein,mainly existing as inclusion bodies,was nearly 60%of total cellular protein and about 48 kD in molecular weight,being in line with the expected size.
