摘要
以黄瓜品种‘津春2号'为试材,利用双向电泳技术研究了聚丙烯酰胺凝胶pH对黄瓜根系和叶片蛋白质双向电泳图谱的影响。结果表明:pH为8.8时,蛋白点大多集中于胶面的中部;pH为8.2的凝胶使小分子蛋白点过于接近溴酚蓝指示线,可能导致某些小分子蛋白丢失;pH为8.5的凝胶能够得到较多和分离较好的蛋白点,适用于黄瓜植株蛋白质组学的研究。
Cucumis sativus cultivar 'Jinchun 2' was used as a test material,and the effects of polyacrylamide gel pH on two-dimensional electropherograms of proteins of cucumber roots and leaves were studied by two-dimensional electrophoretic technique.The results showed that when the pH was 8.8,most of the proteins gathered on the middle of the gel;When the pH was 8.2,the small molecule proteins were excessively close to the bromophenol blue band,possibly causing some small molecule proteins to lose;And when the pH was 8.5,the proteins could be separated more and better, which was suitable for study on cucumber seedlings in proteomics.
