胃癌中SPARC、TGFβ1和Ki-67的表达及意义 被引量:5

Relationship between expression of SPARC and TGFβ1 with cancer cells proliferation in gastric cancer

摘要 目的探讨胃癌组织中SPARC和TGFβ1的表达及其与胃癌细胞增殖的关系。方法用免疫组织化学方法检测80例胃癌组织和30例癌旁正常组织中SPARC、TGFβ1和Ki-67的表达。结果 (1)SPARC在胃癌中的阳性率高于正常胃黏膜(P=0.001)并主要表达于胃癌间质纤维母细胞的胞质,且与胃癌的分化程度、临床分期、Lauren分型及淋巴结转移相关有显著性(P<0.05);(2)胃癌中TGFβ1的阳性率和Ki-67标记指数均高于正常胃黏膜(P<0.001),且均与浸润深度、临床分期和淋巴结转移相关(P<0.05);(3)随着胃癌组织中SPARC的减少及TGFβ1的增多,Ki-67标记指数逐渐增大,且差异有显著性(P<0.05)。结论胃癌组织中SPARC抑制癌细胞的生长,而TGFβ1则促进癌细胞的生长,它们在胃癌的发生、发展中起着重要的作用。 Purpose To study the expression of SPARC and TGFβ1 in gastric cancer and their relationship with cancer cells proliferation.Methods The expression of SPARC,TGFβ1 and Ki-67 was detected in 80 cases of gastric carcinoma and 30 cases of normal gastric tissue by immunohistochemistry.Results(1)The expression of SPARC was significantly higher in gastric cancer than that in normal gastric tissue(P=0.001).The expression of SPARC was mainly found in the tumor associated fibroblasts,and was related to the differentiatial degree,clinical stage,Lauren classification and lymph node metastasis(P0.05);(2)There were statistically significant differences in the expression of TGFβ1 and Ki-67 between gastric cancer and normal gastric tissue(P0.001,respectively).Both the expression of TGFβ1 and Ki-67 in gastric cancer were associated with depth of tumor invasion,clinical stage and lymph node metastasis(P0.05);(3)Ki-67 was positively associated with TGFβ1 but negatively with SPARC in gastric cancer.Conclusion The expression of SPARC inhibits cancer cells proliferation but TGFβ1 promotes cancer cells proliferation in gastric cancer.Both of them are important in the development and progression of gastric cancer.
