SOPC的合成及其在色氨酸酶活力测定中的应用

摘要 以邻硝基苯胺为起始原料,经邻硝基苯胺的重氮化、L-半胱氨酸与邻硝基苯重氮盐的取代反应等步骤制备SOPC,通过活性炭吸附、氨水洗脱和硅胶柱层析纯化,所得产品在熔点、晶形、紫外和红外吸收等方面与文献报道相一致.将合成的SOPC应用于8 株工程菌和1 株宿主菌的色氨酸酶活力测定,结果表明工程菌色氨酸酶的活力比宿主菌均有不同程度的提高,其中WW-53 号比宿主菌高15 倍. S O Nitrophenyl L cysteine was synthesized by dizo reaction of o nitroaniline as start material, replacement reaction of L cysteine and o nitroaniline diazo salt. The product in the reaction mixture was adsorbed with active charcoal, eluted with ammonia solution and purified by silica gel column chromatography. It was corresponding to that reported by other authors, in respects of melting point, crystal shape, UV and IR spectra. The activities of tryptophanase from eight genetic engineering strains and one host strain were determined by synthesized SOPC as substrate. The results showed that the activities of tryptophanase from genetic engineering strains are greater than that of host strain to different extent, and the enzyme activity of No. 53 is of 15 times as high as that of the host.
