坐骨神经慢性结扎损伤模型大鼠脊髓细胞因子表达的变化 被引量:3

摘要 目的研究大鼠坐骨神经慢性结扎损伤(CCI)后脊髓部分细胞因子mRNA表达的变化。方法大鼠随机分为三组:正常组8只作为对照;损伤组32只,结扎左侧坐骨神经制作CCI模型,再按损伤后的第1、3、7、14天四个时点随机均分大鼠,测定痛觉及收集脊髓标本;假损伤组32只,仅暴露左侧坐骨神经不结扎,并同样按四个时点均分大鼠测定痛觉及收集脊髓标本。测定各组脊髓肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β、IL-6和IL-10 mRNA表达情况。结果损伤组大鼠在神经损伤后出现痛觉异常,损伤后第7天最为明显(P<0.05);损伤后第1、3天损伤组TNF-α、IL-1β、IL-6、IL-10 mRNA的表达均显著高于正常组和假损伤组(P<0.05),损伤后第7、14天IL-1β、IL-6、IL-10 mRNA的表达仍明显高于正常组和假损伤组(P<0.05)。结论在大鼠坐骨神经慢性结扎损伤的神经病理痛模型中,脊髓细胞因子的变化可能对病理性疼痛的持续状态起重要的作用。 Objective To observe some spinal cytokines expression in chronic constriction injury of the sciatic nerve in the rat. Methods The rats were randomly divided into three groups. Control group(n= 8). Injured group (the left sciatic nerve was ligated). Sham group (subjected to expose the left sciatic nerves only). The spinal mRNA expressions of TNF-α, IL-11], IL-6 and IL-10 were determined in control group and 1^st, 3^rd, 7^th, 14^th day after surgery in injured group and sham group. Results In the injured group, the allodynia was observed after nerve injury, especially on the 7th day(P〈0. 05). TNF-α, IL-1β, IL-6, and IL-10 were expressed more significant at 1^st and 3^rd day and IL-1β, IL-6, and IL-10 were expressed more significant at 7th and 14th day in the injured group. Conclusion The changes of spinal cytokines may play an important role in the maintenance of neuropathic pain in rat chronic constriction injury model.
