摘要
选取优质马铃薯顶芽作外植体,系统研究了马铃薯茎尖脱毒、壮苗及一步成苗生产技术。结果表明:带菌少的外植体,污染率低,茎尖成活率高;在MS培养基中加入0.2~0.8 mg/L CCC(矮壮素)对脱毒马铃薯试管苗兼有促进增殖、健壮,提高增殖系数,以及促发继代后生根的多重效果;而且试管苗移栽后缓苗期短,生长旺盛。采用一步成苗技术大规模生产,可以简化组培程序,缩短育苗周期,降低生产成本。
The quality apical buds of potato were selected as explants to study the techniques of shoot tip detoxification,vigorous tube-seedlings and one-step seedling culture for potato.The results showed that the less bacterium the explants carried,the lower the contamination rate and the higher the shoot tip survival rate was;adding 0.2~0.8 mg/L of CCC to the MS culture medium had multiple effects for virus-free potato tube-seedlings,which included promoting proliferation and vigorous growth of seedlings,enhancing proliferation coefficient and rooting after subculture;moreover,the tube-seedlings had short temporary planting period and vigorous growth after transplanting.Applying technique of one-step seedling culture to large scale production can simplify process of tissue culture,shorten cycle of seedling breeding,and reducing cost of production.
