史氏鲟肠道微生物基因组DNA提取方法的研究

Extraction Method of Genomic DNA from Intestinal Microorganism of Acipenser schrenki Brandt

以史氏鲟肠道内容物及鲜活肠道为样本,用PBS多次洗涤,离心样品,沉淀菌体。提取肠道菌群总DNA,电泳结果显示,样品DNA条带可用于后续分子生物学试验研究。通过细菌通用引物,对其16S rDNA基因进行PCR扩增,得到较清晰的图谱,条带整齐,表明采用该方法可以用于提取鱼类肠道微生物群落的DNA,且简单、准确、可行。

Taking the intestinal substance and lively intestinal tract of Acipenser schrenki Brandt as sample to precipitate bacteria through PBS cleaning and centrifuge.Total DNA of intestinal bacteria was extracted to investigate intestinal microflora.The results of electrophoresis implied that the banding-patterns of different samples can be used for the following molecular biological experiment.The 16S rDNA in these DNA samples was amplified by PCR with universal primers of bacteria,the clear atlas and the neatly bands were obtained.It is indicated that using this method to extract DNA of intestinal microflora from fish is simple,accurate and feasible.
