摘要
[目的]采用反相高效液相色谱法分析嘧肽霉素发酵液,外标法测定其中抗真菌组分的含量。[方法]使用DIKMA DiamonsilⅡC18反相柱和紫外可变波长检测器,以甲醇-水(体积比为20∶80)为流动相,流速为1 mL/min,检测波长为278 nm。[结果]嘧肽霉素抗真菌组分保留时间为10.2 min,方法的线性相关系数为0.999 8,样品平均回收率为99.4%,变异系数为0.25%。[结论]该方法是用于发酵控制和用药指导的理想分析方法。
An external standard method was developed for the determination of antifungal ingredient in cytosinpeptidemycin fermentation broth by HPLC.[Methods] DIKMA DiamonsilⅡ C18 column and UV detector were used and methanol-water(20∶80,by vol) was mobile phase with flow rate at 1 mL/min and UV detection at 278 nm.[Results] The retention time of antifungal ingredient in cytosinpeptidemycin was 10.2 min.The liner correlation of this method was 0.9998.The average recovery rate and coefficient of variation were 99.4 and 0.25%,respectively.[Conclusions] This is an ideal way for fermentation control and strengthening the guidance on rational drug use.
