肝纤维化大鼠SGK1的表达与肝星状细胞活化的关系及坎地沙坦对SGK1的影响 被引量:1

摘要 目的:观察血清和糖皮质激素调节蛋白激酶1(SGK 1)的表达与肝星状细胞(HSC)活化的关系,并探讨选择性血管紧张素Ⅱ受体亚型AT l拮抗剂坎地沙坦对SGK 1的影响。方法:将36只W istar大鼠随机分为3组:正常对照组、肝纤维化模型组和坎地沙坦治疗组。采用复合法复制肝纤维化模型,坎地沙坦治疗组于第7周初时用大鼠灌胃针灌注药物,实验于10周末全部结束。苏木精-伊红染色法(HE法)和马松三色染色法(M asson法)观察肝组织病理学改变,应用免疫组织化学法检测α-平滑肌肌动蛋白(-αSM A)和SGK 1在肝内的表达及定位,并应用图像分析系统对其阳性表达结果进行半定量分析。结果:各模型组-αSM A,SGK 1的表达量较正常对照组明显增多(P<0.01);坎地沙坦治疗组-αSM A和SGK 1的表达量较同期纤维化模型组明显减少(P<0.0 5)。统计结果表明二者呈正相关(r=0.899,P<0.001)。结论:SGK 1参与了肝星状细胞的激活,坎地沙坦可以抑制肝纤维化的发展,其机制可能与HSC活化受抑、SGK 1表达减少有关。
