摘要
目的:探讨建立腹泻型肠易激综合征(D-IBS)内脏高敏感模型的方法。方法:采用心理应激联合腹腔注射卵清蛋白法诱导IBS大鼠模型,检测其肠道运动(结肠排便时间、排便颗粒)、内脏感觉(腹壁收缩反射、腹壁肌电活动)及结肠黏膜组织学改变。结果:与正常对照组比较,心理应激联合腹腔注射卵清蛋白法可诱导大鼠肠道动力紊乱,内脏敏感性增高(P<0.05,P<0.01),组织学检查无明显炎症性表现。结论:心理应激联合腹腔注射卵清蛋白法建立IBS模型,符合其动力、感觉及组织学特征,可用于IBS的实验研究。
Objective:To establish a rat model of visceral hypersensitivity in diarrhea-type irritable bowel syndrome(D-IBS)by stress induction and intraperitoneal injection of ovalbumin.Methods:Stress and ovalbumin were given to Sprague Dawley rats.After modeling,the colon transit was observed,the rectal distention was performed,the thresholds of abdominal withdrawal reflex were measured,and the abdominal wall myoelectrical activity was recorded.Histology in the colon mucosa was examined.Results:Compared to the control group,the movement of the colon was abnormal,and the thresholds of abdominal withdrawal reflex(P0.05),and the myoelectrical activity(P0.01)were increased in the IBS group.No obvious inflammatory change was noted in histological examination.Conclusion:Stress combined with intraperitoneal injection of ovalbumin can successfully induce D-IBS in rats.This model has the characteristics of abnormal intestinal transit,hypersensitivity,and histology of IBS,which is suitable for experimental studies.
