摘要
目的建立大鼠血浆中儿茶素主要代谢产物没食子酸含量检测方法。方法采用改进的高效液相色谱法测定对大鼠口灌儿茶素后血浆中没食子酸含量,色谱柱为大连依利特BDS C18(250×4.6mm)色谱柱,流动相为乙腈∶0.1%柠檬酸水溶液=5∶95;柱温30℃,波长280nm,以茶碱作为内标。结果改进的HPLC方法测定儿茶素体内代谢产物没食子酸在0.01~20.0mg.L-1范围内线性关系良好(r〉0.999),平均回收率85%以上,精密度10%以下。大鼠口灌儿茶素后血浆中检测到代谢产物没食子酸。结论本研究建立一种灵敏、准确、可靠的RP-HPLC儿茶素代谢产物没食子酸的定量检测方法,并将其应用于大鼠体内的儿茶素代谢研究。
OBJECTIVE To determine the content of galic acid in rat plasma by RP-HPLC.METHODS The contents of galic acid were measured by improved HPLC.Theine was used as internal standard.The determination was performed on SHIMADZU HPLC-20AT Hypersil BDS C18(250×4.6mm) column with acetonitrile-0.1% citric acid(5∶95) as mobile phase at detection wavelength of 280nm.And the column temperature was set at 30℃.RESULTS The linear range of galic acid was 0.01~20.0mg·L-1(r0.999).And these average recoveries were more than 85% and the RSD were less than 10%.The method was used to determine the content of galic acid in rat plasma after oral administration.CONCLUSION A specific,simple,sensitive and accurate RP-HPLC method has been developed and validated for the quantitative determination of galic acid in rat plasma.The method has been applied to metabolic study of catechins in rat.
