摘要
目的 观察犬急性心肌缺血—再灌注损伤的早期酶组织化学改变。方法 复制犬急性心肌缺血—再灌注损伤动物模型,观察糖原磷酸化酶、分枝酶、琥珀酸脱氢酶、乳酸脱氢酶、β羟丁酸脱氢酶、细胞色素氧化酶、腺苷三磷酸酶活性变化。结果 缺血30 min ,糖原磷酸化酶和分枝酶呈阴性染色,琥珀酸脱氢酶、乳酸脱氢酶、β羟丁酸脱氢酶、细胞色素氧化酶和腺苷三磷酸酶活性轻度降低。再灌注120 min ,糖原磷酸化酶和分枝酶活性略有恢复,其它酶无明显改变。缺血60 ~90 min ,琥珀酸脱氢酶、乳酸脱氢酶、β羟丁酸脱氢酶、细胞色素氧化酶和腺苷三磷酸酶活性进一步降低,再灌注120 min ,缺血区扩大。结论 再灌注可使早期缺血心肌恢复,酶组织化学染色法有助于急性心肌缺血再灌注损伤的诊断。
Objective To observe the enzyme histochemical changes of coronary occlusion followed by reperfusion in dog.Methods The model of ischemic reperfusion was established in dog.The activities of PPr,BE,SDH,LDH, β HBDH,CCO and ATPase were observed with enzyme histochemical technique.Results At 30 min of ischemia,the activities of PPr and BE were negative ,the activity of other enzymes reduced slightly.After 120 reperfusion,the activities of PPr and BE were restored a little,there were no significant changes in the activity of the other enzymes.At 60 to 90 min,ischemia,the activities of SDH,LDH,β HBDH,CCO and ATPase were diminished apparently.After 120 min.reperfusion,the PPr and BE were still negative while the area of ischemia stained by the other enzymes was enlarged.Conclusion The resultes show that early ischemic myocardium can be salvaged by reperfusion and enzyme histochemical stain is helpful for the diagnosis of acute myocatdial ischemia followed by reperfusion.
