摘要
目的:建立比格犬血浆中喹硫平浓度测定的HPLC-MS/MS定量分析方法,研究喹硫平缓释片在犬体内的药动学特征。方法:采用6只健康成年比格犬随机分成2组,双交叉试验设计进行单剂量给药,并于相应的时间点采集血样,采用HPLC-MS/MS测定比格犬体内的血药浓度并计算主要药动学参数。结果:经统计分析,与普通参比制剂相比,受试缓释制剂的Cmax降低,tmax延迟,t1/2延长。结论:缓释片与普通片相比具有明显的缓释特征。
Objective: To establish an HPLC-MS/MS method for the determination and pharmacokinetic study of quetiapine in Beagle dogs.Methods: Six beagle dogs were randomized into two groups.A two-way crossover study was adopted.Plasma samples were collected at different time points.Plasma concentration of quetiapine was determined by HPLC-MS/MS and pharmacokinetic parameters were calculated.Results: The results indicated that the tmax and t1/2 of extended-release tablets were longer than those of immediate tablets,and the Cmax of extended-release tablets was lower than that of immediate tablets.Conclusion: The test tablets have marked sustained release characters.
