摘要 目的探讨国产137Csγ射线血液辐照机辐照对悬浮红细胞中淋巴细胞增殖能力的影响,以及辐照后悬浮红细胞在保存期间质量的变化。方法将悬浮红细胞分为未辐照组及辐照组,辐照组使用国产137Csγ射线血液辐照机进行照射,照射剂量为25 Gy。分离辐照前后悬浮红细胞中的淋巴细胞进行培养,观察辐照前后淋巴细胞增殖率的变化;检测辐照后保存0、5、10、20 d的悬浮红细胞的pH值、电解质浓度、红细胞计数、红细胞压积(Hct)、平均红细胞体积(MCV)、血小板计数(PLT)、平均血小板体积(MPV)以及游离血红蛋白浓度,并与未辐照组进行比较。结果未辐照组淋巴细胞增殖率为(99.64±11.63)%,辐照组淋巴细胞增殖率为(0.98±0.22)%,两组比较差异有统计学意义(P<0.05)。辐照后的悬浮红细胞保存20 d Na+、K+、游离血红蛋白浓度与未辐照组比较,差异有统计学意义(P<0.05)。结论国产137Csγ射线血液辐照机采用25 Gy剂量辐照悬浮红细胞,可有效杀灭淋巴细胞,预防输血相关性移植物抗宿主病。 Objective To investigate the effect on the proliferation of lymphocytes of suspended red blood cells irradiated by homebred 137Cs γrays blood irradiator,and to explore the quality changes of suspended red blood cells during preservation after irradiation.Methods The suspended red blood cells were divided into irradiation group and non-irradiation group.The irradiation group was irradiated by 137Cs γ rays blood irradiator with a dose of 25 Gy.The lymphocytes were isolated from the suspended red blood cells suspension before and after irradiation,and were cultured so as to observe the changes of cellular proliferation rate.PH,electrolytes concentration,RBC counting and Hct,MCV,PLT,MPV and FreeHb in both groups were detected and compared on the 0,5 th,10 th,and 20 th day.Results The rate of lymphocytes proliferation in non-irradiation group was(99.64±11.63)%,but(0.98±0.22)% in irradiation group,and there was significant difference between the two groups(P0.05).There were significant differences in Na+,K+,FreeHb between the two groups(P0.05).Conclusion The homebred 137Cs γ rays blood irradiator irradiating suspended red blood cells with a dose of 25 Gy can kill lymphocyte and prevent TA-GVHD effectively.
